| The small-scale grouper Epinephelus polylepis from the family Epinephilidae, is an important commercial species that plays a crucial role in coastal benthic ecosystems. Previous studies have failed to accurately identify this species in the northern region of the Persian Gulf and the Gulf of Oman. The aim of this study is to analyze the samples of E. polylepis collected in the Persian Gulf and Gulf of Oman using a combination of morphological and molecular data. The morphological data include 17 morphological characters and six counting characters. For the molecular study, sequences of the mitochondrial COI were used, which were combined with GenBank sequences. The samples were collected in the sandy areas of two stations, Tis and Qeshm. This species can be distinguished from other congeners by the presence of smaller and closer spots on the fins and spots on the back of the head and body. Analysis of the COI sequences of the mitochondrial DNA and samples sequenced in this study and samples from the vicinity of the typing site (Bahrain) revealed a monophyletic group with a high posterior probability and a low intraspecific genetic distance (0.19%). مقدمه خانوادهEpinephilidae (راسته سوف ماهی شکلان) شامل گونههای تجاری مهم است (Luiz et al., 2016). این خانواده قبلاً بهعنوان زیرخانواده Epinephelinae در خانواده Serranidae قرار داشته است که بهتازگی از خانواده Serranidae جدا و به سطح خانواده Epinephilidae ارتقا پیدا کرده است (Smith & Craig, 2007 Ma & Craig, 2018; Frick et al., 2024; ). گروپرها معمولاً با شکل بدن و سر، الگوهای رنگی، و همچنین اندازه عناصر باله شناسایی میشوند (Heemstra & Randall, 1993; Randall, 1995) که ریختشناسی همگن آنها میتواند باعث شناسایی نادرست گونههای نزدیک شود (Rimmer & Glamuzina, 2019 Hassanien & Al-Rashada, 2021; ). این خانواده شامل 16 جنس و 170 گونه در نواحی گرمسیری و نیمهگرمسیری است که 4 جنس و 15 گونه آن در خلیج فارس و خلیج عمان پراکنش دارند (Fricke et al., 2024). جنس Epinephelus با 91 گونه، بیشترین تنوع گونهای را بین هم جنسهای خود دارد و از اقیانوس اطلس تا اقیانوس هند-آرام شامل خلیج فارس و خلیج عمان پراکنش دارد (Fricke et al., 2024). در گذشته مطالعه کمی در ارتباط با این جنس مهم در خلیج فارس و خلیج عمان انجام گرفته است. مطالعات برجسته در ارتباط با گونههای این جنس شامل چکلیستها (Blegvad, 1994; Carpenter et al., 1997)، بررسی اتولیت شش گونه (E. Bleekeri، E. coioides، E. dicanthus، E. stoliczkae،latifasciatus E. areolatus E.) (Javadzadeh et al., 2017 Edalati et al., 2018; )، چکلیست ماهیان خلیج فارس ازجمله جنس Ephinephelus (Eagderi et al., 2018) و بررسی مولکولی گونه E. coioides (Tavakoli-Kolour et al., 2022) انجام گرفته است. گونههای این جنس نقش بومشناختی مهمی در زیستگاههای ساحلی و فراساحلی ایفا میکنند؛ زیرا این گونهها معمولاً شکارچیان برتر در اکوسیستم کفی هستند (Moazzam & Osmany, 2023). گروپرها معمولاً با شکل بدن و سر، الگوهای رنگی و همچنین اندازه باله شناسایی میشوند (Heemstra & Randall, 1993) که شباهت ریختشناسی آنها میتواند باعث شناسایی نادرست بین گونهها در میان گونههای خاصی از جنس Epinephelus ازجمله گروپر فلس کوچک شود. گروپر فلس کوچک Epinephelus polylepis با پراکندگی گسترده در اقیانوس هند غربی یکی از منابع اولیه صید ماهی با ارزش در خلیج فارس و خلیج عمان شناخته میشود (Fricke et al., 2024). این گونه اولینبار از بحرین در خلیج فارس توسطRandall and Heemstra (1991) توصیف و نامگذاری شد. با توجه به شباهت ریختی، Epinephelus polylepis با گونههای دیگر در جنس Epinephelus ازجمله E. chlorostigma، E. bleekeri، E. lanceolatus ،E. areolatus اشتباه گرفته میشود. استفاده از دادههای مولکولی در مطالعات ردهبندی جانوران مختلف در سالهای اخیر افزایش یافته است (Hebert et al., 2004). در مطالعات مولکولی توالی بخشی از زیر واحد سیتوکروم اکسیداز I (COI) برای شناسایی در سطح گونه (Asgharian et al., 2011; Damadi et al., 2020; Alavi-Yeganeh et al., 2021; Mehraban et al., 2021; Damadi et al., 2023) و آنالیزهای ریختشناسی ( Khayyami et al., 2015 Damadi et al., 2021;) و همچنین، ریختسنجی هندسی (Polgar et al., 2017 Ghanbarifardi et al., 2020; Sadeghi et al., 2020; Damadi et al., 2024) در خلیج فارس و خلیج عمان استفاده میشود. هدف این مطالعه، شناسایی گونه E. polylepis با رویکرد ریختشناسی و مولکولی با ژن COI برای اولینبار در خلیج فارس و خلیج عمان است. مواد و روشها طی مطالعه میدانی بین سالهای 1400 تا 1401، نمونههای متعلق به E. polylepis با استفاده از تورهای آبششی و قلاب جمعآوری شدند. این نمونهها به موزه جانورشناسی دانشگاه فردوسی مشهد (ZMFUM) منتقل شدند. نمونهها ازنظر شکل، رنگ و سایر صفات اندازشی و شمارشی بررسی شدند. نمونهها از سمت چپ عکسبرداری شدند. سپس، بخش کوچکی از بافت عضلانی از هر نمونه تازه جدا شد و در اتانول مطلق نگهداری شدند. سپس کل نمونهها در فرمالین 10 درصد تثبیت شدند. مطالعات ریختسنجی صفات ریختشناسی براساس منابع موجود ازجمله جانسون و همکاران (Johnson et al., 2019) استخراج و بررسی شدند. نمونهها با کلیدهای شناسایی معتبر (Randall, 1995 Johnson et al., 2019; ) شناسایی شدند. درمجموع از 23 صفت ریختشناسی شامل 17 صفت اندازشی و شش صفت شمارشی استفاده شد (جدول 1). صفات اندازشی و شمارشی به ترتیب با استفاده از کولیس دیجیتالی با دقت 01/0 و استرئومیکروسکوپ مدل (Olympus SZ40) بررسی شدند. برای حذف اثر اندازه از صفات ریختسنجی نسبت گرفته شد.
مطالعات مولکولی استخراج DNA از سه نمونه با استفاده از روش نمکی انجام گرفت (Bruford et al., 1992). تکثیر قطعه ژنی با پرایمرهای FishF1(5' (TCAACYAATCAYAAAGATATYGGCAC 3' و (5' ACTTCYGGGTGRCCRAARAATCA 3') FishR1 صورت گرفت. برنامه دمایی برای ژن COI بهصورت حرارت 95 درجه سانتیگراد در مدت 5 دقیقه، با 35 سیکل چرخه حرارتی ، 45 ثانیه دناتوره در دمای 94 درجه، دمای اتصال 45 ثانیه در 52 درجه، مرحله طویلشدن 90 ثانیه در دمای 72 درجه دنبال شد و درنهایت، به مدت 7 دقیقه در دمای 72 درجه سانتیگراد پایان پذیرفت. کیفیت نهایی محصول با الکتروفورز ژل آگارز بررسی شد. الکتروفورز روی ژل آگارز 5/0درصد و با رنگآمیزی Green viewer با ولتاژ 85 ولت برای حرکت DNA استفاده گردید و در انتها از دستگاه مستندساز ژل برای مشاهده باندهای DNA استفاده شد. محصولات حاصل از PCR برای تعیین توالی به شرکت میکروسینس سوئیس ارسال شد. ترابهای این مطالعه با توالیهای حاصل از بانک ژنی (GenBank) ترکیب شد. برون گروه براساس مطالعه ما و همکاران (Ma et al., 2018) انتخاب شد. تراتبهای ژن میتوکندریایی با نرمافزار (Hall, 1999) BioEdit v7.2 تصحیح و با نرمافزار (Kumar et al., 2018) MEGA v10.0 هم تراز شدند. برای بررسی کدونهای متوقفکننده، توالیهای کدکننده پروتئین به توالیهای آمینواسیدی ترجمه شدند تا در صورت حضور آنها از مجموعه توالیها حذف شوند. فاصله ژنتیکی درون گونهای و بین گونهای که درنتیجه تفاوت نوکلئوتیدی است، با مدل (K2P)Kimura Two-Parameter و با نرمافزار (Kumar et al., 2018) MEGA v10.0 محاسبه شد. بهترین مدل جایگزینی تکاملی برای نوکلئوتیدها با استفاده از (Darriba et al., 2012) jModelTest v2.1.6 و با توجه به معیار Akaike Information criterion شناسایی شد. تحلیل بیژین با نرمافزار (Ronquist et al., 2012) MrBayes 3.2.7 و تعداد 30000000 تکرار انجام شد. در تحلیل بیژین 20 درصد درخت اولیه از تحلیل حذف شد و پارسیمونیترین درخت حاصل از بیژین با نرمافزار (Rambaut, 2016) FigTree v1.4.4 ویرایش شد. کلادها با احتمال پسین بالای 95 بهعنوان کلادهای تأییدی در نظر گرفته شدند. نتایج از دو ایستگاه تیس (خلیج عمان) (سه نمونه) (25°33'99''N; 60°60'02''E) از عمق 30 تا 50 متری و قشم (خلیج فارس) (دو نمونه) از عمق 25 تا 30 متری (26°58'75''N; 55°48'34''E) (شکل ۱) درمجموع، 5 نمونه از گونه Epinephelus polylepis (شکل ۲) با تور و قلاب ماهیگیری جمعآوری شد.

دادههای ریختشناسی صفات ریختشناسی: ویژگیهای ریختشناسی در گروپر فلس کوچک (شکل 2) شامل: 11 خار (XI) و 17-16 شعاع نرم در باله پشتی؛ 70-66 فلس در امتداد خط جانبی؛ 8 شعاع نرم در باله مخرجی؛ 19 شعاع نرم در باله سینهای؛ 10-9 خار آبششی در نیمه بالایی کمان اول و 17-18 خار آبششی در نیمه پایینی کمان اول (25-28 مجموع خارهای آبششی)؛ باله دمی کوتاه یا کمی لبهدار؛ سرپوش آبششی دندانه دار؛ سر، بدن و بالههای با تعداد زیادی لکههای قهوهای تیره کوچک و نزدیک پوشیدهشده (به جز قسمتهای شکمی سر و بدن)؛ لبه انتهایی باله دمی با خط سفید و یک ردیف لکههای قهوهای مایل به سیاه؛ خطوط تیره در فک بالا.
آنالیز مولکولی درمجموع، 65 توالی متعلق به 57 گونه از جنسEpinephelus و یک برون گروه Cephalopholis boenak مربوط به ژن میتوکندریایی سیتوکروم C اکسیداز زیرواحد (COI) Iبرای تحلیلهای مولکولی استفاده شدند. پس از همترازی توالیهای نهایی گونههای این جنس تعداد 654 نوکلئوتید برای تحلیلهای استفاده شد که شامل 180 نوکلئوتید پلی مورف، 282 نوکلئوتید محافظتشده و 167 نوکلئوتید پارسیمونی بود. مدل GTR+I+G بهعنوان بهترین مدل انتخاب شد. جنسEpinephelus با توجه به حضور گونه Anyperodon leucogrammicus پارافیلی بود و در تجزیهوتحلیل بیژین جنسEpinephelus به پنج کلاد مجزا با حمایت متوسط تا بالا تقسیم شده است ((A-E (شکل 3). گونهEpinephelus polylepis ارتباط بسیار نزدیکی با E. areolatus، E. cyanopodus و E. multinotatus بهعنوان گروه خواهری را نشان داد (شکل 3). همچنین، در آنالیزهای تبارزادی، نمونههای توالییابیشده در این مطالعه با نمونههایی از نزدیک محل تایپ (بحرین) یک گروه تک نیا با احتمال پسین بالا را شکل دادند (شکل 3). تفاوت ژنتیکی بین نمونههای E. polylepis و گروه خواهری آنها به ترتیب E. areolatus (3/8 درصد) ، E. bleekeri (8/11 درصد) و E. chlorostigma (6/10 درصد) بود؛ درحالیکه میانگین فاصله ژنتیکی درون گونهای آنها 0.19 درصد است.

The sequences of Epinephelus polylepis studied in this study are shown in red. بحث مطالعه حاضر شناسایی گونه Epinephelus polylepis با رویکرد یگانشی از خلیج فارس و خلیج عمان را نشان میدهد که قبلاً مطالعه جامعی صورت نگرفته بود. بسیاری از گونههای دریایی ازجمله گونههای گروپر به دلیل شباهت زیادی که دارند، سبب شناسایی نادرست یا گزارشهای گمراهکننده از گونهها میشوند (Heemstra & Randall, 1993). چنین مواردی میتواند منجر به عدم دقت دادهها برای شیلات و همچنین، ارزیابی اشتباه در ارتباط با تنوع گونهای مدنظر شود؛ بنابراین، مشکل شناسایی گونههای مختلفEpinephilidae میتواند در تمام مناطق ساحلی اقیانوس هند رخ دهد (Craig et al., 2001 Tavakoli-Kolour et al., 2022; ). تنوع بسیار بالا در سطح گونه، نبود تفاوتهای ریختشناسی مشخص و تخصص ناکافی در ردهبندی گونههای گروپر سه عاملی هستند که احتمال شناسایی نادرست گونه و درنتیجه، برداشت بیش از حد احتمالی گونههای ناخواسته را افزایش میدهند (Craig et al., 2001 Tavakoli-Kolour et al., 2022; ). روابط تبارزادی بین اعضای جنس Epinephelus مدتهاست که بهخوبی درک نشده و با نمونههای محدود مطالعه شده است (Craig & Hastings, 2007 Ma & Craig, 2018; ). در این مطالعه، روابط تبارزادی بیش از 60 درصد گونههای جنسEpinephelus (57 گونه از 89 گونه) بهعنوان ابزاری برای ایجاد یک فرضیه تبارزادی با استفاده از تحلیل بیژن و بیشینه درستنمایی بررسی شد؛ درحالیکه نتایج ریختشناسی، تک نیایی دو جنس Anyperodon و Epinephelus را در گذشته تأیید کرده است ( Johnson, 1983; Leis, 1986)؛ با این حال نتایج مولکولی ما همراه با مطالعات قبلی (Ma et al., 2018)، جنس Günther 1859 Anyperodon شامل گونه تایپ Anyperodon leucogrammicus را در جنس Epinephelus Bloch 1793 قرار میدهد و Anyperodon احتمالاً باید بهعنوان مترادف Epinephelus در نظر گرفته شود. علاوه بر این، کلادهای حاصله در جنس Epinephelus ازنظر الگوی رنگی و بومشناختی باهم تفاوت دارند (Ma et al., 2018). هر دو داده مولکولی و ریختشناسی اعتبار E. polylepis را با حمایت بالا پشتیبانی میکنند (شکل 3، جدول1). این گونه از غرب سواحل هند تا یمن گزارش شده است و در گذشته به اشتباه بهعنوان E. chlorostigma که یک گونه پیچیده (species complex) شناخته شده است (Heemstra & Randall, 1993). همه ویژگیهای شمارشی و الگوی رنگی نمونههای جمعآوریشده این مطالعه تا حد زیادی با نمونههای تایپ و پاراتایپ همپوشانی دارند (Randall & Heemstra, 1991 Heemstra & Randall, 1993; ). مقایسه این نمونهها دربارۀ ویژگیهای ریختسنجی نیز با نمونه اصلی سازگار بود، بهجز دو صفت عمق بدن و طول سر که در نمونههای ما تا حدی بزرگتر است. این تغییرات میتواند به دلیل نحوه اندازهگیری متفاوت توسط نویسندگان در این دو مطالعه انجام گرفته باشد. همچنین، ویژگیهای توصیفی نمونههای گزارششده از عمان ((Randall, 1995، پاکستان (Psomadakis et al., 2015 Moazzam & Osmany, 2023; ) و شمال غرب اقیانوس هند (Heemstra et al., 2022) نمونههای بررسیشده در این مطالعه را تأیید میکنند. گونه E. polylepis ازنظر ریختشناسی و مولکولی بیشترین شباهت را با گونههایE. areolatus ، E. bleekeri و E. chlorostigma در خلیج فارس و خلیج عمان نشان میدهد (شکل 3). گونه E. chlorostigma اکثراً به اشتباه با E. polylepis شناسایی میشود. این دو گونه بهوسیله تعداد فلسهای روی خط جانبی (66-70 در E. polylepis در مقابل 53-48 در E. chlorostigma) و باله مخرجی پهنتر در E. polylepis در حالی که در E. chlorostigma باله مخرجی نوک تیزتر است (Randall, 1995). گونه E. polylepis از E. areolatus و E. bleekeri بهوسیله تعداد خارهای آبششی (17-18 خار آبششی در نیمه پایینی در مقابل 16-14 خار آبششی در E. areolatus) و الگوی رنگی (لکههای قهوهای تیره کوچک و نزدیک در سطح بدن در مقابل لکههای قهوهای تیره بزرگ و با فاصله زیاد در E. areolatus) جدا میشود. تجزیهوتحلیل مولکولی، شناسایی ریختشناسی نمونههای محدوده شمالی خلیج فارس و خلیج عمان بهعنوان E. polylepis را تأیید کرد (شکل 3). در آنالیز تبارزادی توالی میتوکندری COI یک گروه تک نیا شامل نمونههای محدوده شمالی خلیج فارس و خلیج عمان و نمونههای بحرین (مکان تایپ) را ثابت کرد که واگرایی کمی بین آنها وجود دارد. تجزیهوتحلیل ژنتیکی نیز واگرایی بالایی بین E. polylepis و گونههای نزدیک ازجمله E. chlorostigma و E. areolatus نشان داد. اگرچه روابط تبارزادی این گونهها را نمیتوان تنها با ژن COI بهخوبی حل کرد، هر یک از گونهها یک کلاد تک نیا را با پشتیبانی بالا تشکیل دادند که از اعتبار آنها حمایت میکند (شکل 3). نتایج فاصله ژنتیکی مطالعه حاضر نیز جدایی بین E. polylepis و سه گونه نزدیک E. areolatus ، E. bleekeri و E. chlorostigma را بیش از حداقل فاصله تعیینشده شناسایی کرد (3/8، 8/11 و 6/10 درصد به ترتیب). این فاصله ژنتیکی در ماهیان دریایی براساس ژن COI برای جدایی گونههای نزدیک یا آرایههای خواهری حداقل 2 درصد تعیین شده است (Ward, 2009) که اعتبار این گونه را ازنظر ژنتیکی تأیید میکند. همچنین، براساس دادههای بانک ژن E. polylepis با شباهت 99 تا 100 درصد دربارۀ توالیهای COI با موفقیت بهعنوان E. polylepis شناسایی شد. اگرچه توالیهایی از بانک ژن مشاهده میشود که گونههایE. areolatus ، E. chlorostigma و E. bleekeri را به اشتباه بهعنوان E. polylepis شناسایی کردند (از آنالیز حذف کردیم)، به نظر میرسد شناسایی نادرست گروپر Epinephelus بهطور فراگیر و بین گونهای رخ میدهد. در مطالعات آینده شناسایی دقیق مولکولی دیگر اعضای خانوادهEpinephilidae میتواند روابط و تنوع گونهای آنها را بهخوبی به اثبات برساند.
